I want to upgrade my HD from 40 GB 7200 RPM to about a 100 GB one?
Is it necessary to backup any data? Will I need to reinstall Windows?
Any other tips before I venture forth??

Actually, what I should have asked was: how do I get my programs and data from the old drive to the new one??

Most retail drives come with a program to copy your entire drive's content to the new one. If you're getting it OEM you might be able to get the copy prog. and the vendors website. If not, use Norton Ghost.
do you have a use for the 40g? if not just setup the 100g as a slave. i always recommend keeping windows and programs installed on a smaller drive and just have a nice big drive for storage.
if you simply want to mirror, i found norton ghost to be the best software implementation. i personally prefer doing fresh installs if it's jsut a 1 shot deal tho.
as always, if something is to important to lose, then back it up....you never know.
